Beta-glucans — the standardisation basis

The fungal cell wall is built substantially from beta-glucans: glucose polymers joined by beta-1,3 linkages with beta-1,6 branches. This branched 1,3/1,6 architecture is characteristic of fungi — it differs from the linear 1,3/1,4 beta-glucans of oats and barley — and it is the best-studied class of immune-active fungal polysaccharides. In a hot-water extract of Hericium erinaceus fruiting body, beta-glucan is the dominant genuine active fraction, and a serious commercial extract is standardised on it: our flagship grade runs not less than 30% w/w on a dry basis, with a 40% grade available.

Alpha-glucans — the anti-marker

Alpha-glucan is glucose joined by alpha linkages — which is to say, starch and glycogen. Fruiting bodies contain little of it. Grain contains a great deal. A lion's mane powder with high alpha-glucan is telling you its history: it grew on grain, and the grain came with it. This is why alpha-glucan deserves a limit on the specification (we hold NMT 5%) rather than being left unmeasured.

Hericenones and erinacines — the terpenoid fractions

The compounds most cited in the research literature on this species are not polysaccharides at all. Hericenones are aromatic compounds of the fruiting body; erinacines are diterpenoids concentrated in the mycelium. Both families are studied in nerve-growth-factor (NGF) pathway research — the research rationale behind lion's mane's cognitive positioning. Two practical consequences: a fruiting-body extract legitimately carries hericenones, not erinacines, so an erinacine claim on fruiting-body material is a red flag; and because these compounds are poorly water-soluble, capturing them takes a dual extraction — hot water for the glucans, then a hydroethanolic step for the terpenoids.

Two Methods, Two Very Different Numbers

Enzymatic assay — the method to insist on

The enzymatic method (the Megazyme mushroom beta-glucan kit is the industry reference, built on AOAC-validated chemistry) works by specificity: total glucan is enzymatically hydrolysed to glucose and measured; alpha-glucan is separately hydrolysed with amyloglucosidase and measured; and beta-glucan is the difference. Because the enzymes only cut their own linkage types, starch cannot masquerade as beta-glucan — the method reports it separately, as alpha-glucan. One assay therefore yields both the active figure and the adulteration readout.

Phenol-sulfuric UV — why "polysaccharides 50%" misleads

The phenol-sulfuric method dehydrates carbohydrates in hot sulfuric acid and reads the resulting colour by UV-Vis spectrophotometry. It is fast and cheap, and it responds to every carbohydrate in the sample: beta-glucan, yes, but equally starch from grain substrate, maltodextrin carrier, chitin fragments and free sugars. A myceliated-grain powder that is half starch will post a superb "polysaccharide" number. The figure is not fraudulent in itself — it is simply unable to distinguish the material you want from the material you are trying to avoid.

The key insight: on genuine fruiting-body extract, the UV polysaccharide figure and the enzymatic beta-glucan figure differ moderately. On grain-based or carrier-diluted material, they diverge wildly — 50% "polysaccharides" against single-digit beta-glucan is a real and common pattern. The divergence itself is diagnostic. Ask for both numbers and the alpha-glucan, and the material has nowhere to hide.

The extract-ratio point deserves emphasis because it migrates here from the herb trade: an "8:1" or "10:1" lion's mane says how much raw material went in, not what came out. Starting-material quality drives the result entirely — an 8:1 from myceliated grain is 8:1 concentrated starch. For any product carrying a defined-active claim, you need the enzymatic percentage.

Writing a Specification That Cannot Be Gamed

  1. Name the method. "Beta-glucans NLT 30.0% w/w on dry basis, by enzymatic assay (Megazyme-type)." Without the method clause, a UV polysaccharide figure is a defensible reading of your own specification.
  2. Set the alpha-glucan limit. NMT 5%. This single line excludes myceliated grain from your supply chain more effectively than any declaration.
  3. State the part. "Fruiting body only; no mycelium, no grain substrate" — on the CoA, batch by batch, not on a brochure.
  4. Require carriers declared. By name and percentage. A spray-dried water-soluble grade legitimately carries one; an undeclared carrier is dilution.
  5. State the dry basis. Beta-glucan powders are hygroscopic; without a dry-basis clause, moisture uptake quietly erodes your assay.

What About the Terpenoids?

Hericenone quantification is not yet harmonised across the industry — there is no pharmacopoeial monograph, and inter-lab comparability is poor — which is why we standardise on beta-glucans and report hericenones per batch on the dual-extract grade rather than promising a universal percentage. Treat any supplier quoting a precise hericenone standardisation with the same scrutiny you would apply to the method column generally: ask which reference standard, which method, which laboratory.
