Lion's mane combines a premium price, explosive demand, and a buyer base that mostly checks a single "polysaccharide" number generated by a method that cannot tell mushroom from starch. That is a precise description of the conditions under which adulteration flourishes — and in this category the dominant form is not a spiked or faked material but a legal, cheaper product wearing the wrong label. Each route below has a specific test that catches it.
Why Lion's Mane Is a Target
Botanical adulteration follows economics. Fruiting-body cultivation takes months of controlled growing before harvest; myceliated grain takes weeks in a fermentation room. The finished powders look similar, the cheap one posts a better "polysaccharide" number on the assay most buyers use, and the price difference is large. No malice is required for the market to fill with material that is mostly substrate — only inattention.
The result, in most cases, is not a fake product. It is a real fungal product that is mostly not mushroom.
Route 1 — Mycelium-on-Grain Sold as Mushroom
The signature adulteration of the category, and the reason the fruiting-body question dominates every serious specification discussion. Mycelium is grown on sterilised grain (rice, oats, sorghum); when harvest time comes, the colonised grain block is dried and milled whole, substrate included. The resulting powder is legitimately "Hericium erinaceus biomass" — and substantially starch. Published analyses of commercial "lion's mane" products have repeatedly found grain-based materials with a fraction of the beta-glucan of fruiting-body extract and alpha-glucan contents that no mushroom produces.
What catches it: the enzymatic glucan split. Fruiting-body extract: beta-glucan high (25–40%), alpha-glucan low (under 5%). Myceliated grain: the reverse. One assay, run properly, settles the question per batch. A supporting tell is the classic iodine test — starch stains blue-black with iodine, and a grain-heavy powder responds dramatically while genuine fruiting-body extract barely reacts.
Specification language that works: "Fruiting body only; no mycelium, no grain substrate. Beta-glucans NLT 30.0% w/w by enzymatic assay, on dry basis; alpha-glucans NMT 5.0% w/w by the same method. Carriers and excipients, if any, declared by name and percentage on the Certificate of Analysis." A supplier selling grain biomass now has to either disclose it or falsify a document — a materially different decision.
Route 2 — Starch and Maltodextrin Dilution
The herb-trade classic, applied to mushrooms: genuine extract stretched with maltodextrin, rice flour or dextrose. When declared, a carrier is legitimate — a spray-dried water-soluble grade genuinely needs one. Undeclared, it is dilution, and in this category it hides especially well because the UV polysaccharide assay counts the carrier as active: adding maltodextrin to lion's mane extract can raise the "polysaccharides" figure while gutting the beta-glucan.
What catches it: the same enzymatic split (maltodextrin is alpha-glucan and reports as such), plus a declared-carrier line and a dry-basis clause in the specification.
Route 3 — The Polysaccharide Number Game
Not adulteration of the material but of the arithmetic: selling on "polysaccharides 50% by UV" while staying silent on beta-glucan. As covered in our standardisation guide, the phenol-sulfuric UV method responds to every carbohydrate present — starch, carriers, free sugars — so the figure is unfalsifiable and nearly meaningless. Material that cannot survive an enzymatic assay is marketed exclusively on the UV number.
What catches it: refusing the UV figure as a release criterion. Specify the enzymatic method by name and treat a supplier's reluctance as data.
Route 4 — Extract-Ratio Mislabelling
An "8:1" or "10:1" lion's mane sold into a specification written for a standardised grade, on the logic that a big ratio sounds strong. The ratio describes input weight, not output actives — an 8:1 concentration of myceliated grain is concentrated starch. Often this is sloppiness rather than dishonesty, which does not make the material any better.
What catches it: requiring an assay figure with a named method, and refusing to accept a ratio as a substitute for one.
Route 5 — Species Substitution
Other Hericium species (H. coralloides, H. americanum) and, at the cruder end, cheaper cultivated fungi can be milled into or substituted for H. erinaceus. As dried powder, no visual inspection can tell the difference.
What catches it: HPTLC identity against reference Hericium erinaceus, and — the higher evidentiary standard — ITS DNA barcoding of the incoming raw material. DNA methods work well on raw fruiting body; on a finished hot-water extract DNA is degraded, which is precisely why identity should be locked at the raw-material stage and carried forward by batch traceability. Ergosterol, the characteristic fungal sterol, is a useful supporting marker: it confirms genuine fungal tissue and its level is implausibly low in heavily grain-diluted powders.

Practical Steps for a Buyer
- Specify the enzymatic method by name. The single highest-value sentence in a lion's mane specification.
- Set alpha-glucans at NMT 5%. The one line that structurally excludes grain from your supply chain.
- Get "fruiting body only" on the CoA, not in correspondence. Batch documents are what audits read.
- Require carriers declared by name and percentage. Converts an omission into a falsification.
- Third-party verify the first batch. Nominate Eurofins, SGS or Intertek for pre-shipment testing on a new supplier, and have them run the glucan split, not just the UV number.
- Re-qualify periodically. Supply chains drift, and mushroom supply chains drift faster than most — demand has outrun cultivation capacity for years.
