Standardisation and Identification Are Different Jobs

On a well-behaved botanical, the marker assay implicitly confirms identity: an extract without piperine is not black pepper, an extract without forskolin is not coleus. Assay and identity collapse into one measurement.

Giloy breaks that convenience, for the same structural reason licorice does:

A bitter-principles specification is satisfied by either. The assay is doing its job correctly and telling you nothing about identity.

Why This Matters More Here Than on Licorice

On licorice, buying G. uralensis instead of G. glabra is a commercial and compliance problem — the wrong species, a different flavonoid profile, a registration that no longer matches.

On giloy the consequence is different in kind. Hepatotoxicity concerns are documented for T. crispa, and when liver-injury reports associated with giloy products prompted scrutiny in India, species misidentification was among the explanations proposed by researchers and reviewers.

The practical framing: on giloy, species verification is not part of the quality system — it is part of the safety system. That changes where it sits in a specification, who signs off on it, and whether "the supplier says it's cordifolia" is an acceptable control. It is not.

What Fingerprinting Actually Does

Since no single marker separates the species, the control is the whole chromatogram.

T. cordifolia carries a characteristic constituent profile — including tinosporide, tinocordiside and related clerodane furanoditerpenes and glycosides — in a pattern that differs from T. crispa's. HPLC fingerprinting compares the sample's full profile against authentic reference material rather than checking one peak.

The method in outline:

  1. Establish a reference profile from authenticated T. cordifolia stem.
  2. Run the sample under matched chromatographic conditions.
  3. Compare the pattern — peak presence, relative proportions, retention times.
  4. Report match or mismatch, with T. crispa specifically excluded.

Where a higher evidentiary standard is wanted, DNA barcoding of the incoming raw material is available — more definitive on plant material, though of limited use on a finished extract where DNA is degraded. The practical combination is DNA or botanical authentication at intake plus fingerprinting on the finished extract.

Note the fourth row's caveat. Bitter principles tells you how concentrated the extract is — a genuinely useful thing to know, and the right basis for dosing. It simply is not an identity test, and treating it as one is the error this article exists to prevent.

The Assay's Own Limitations

So state the method, and use berberine by HPLC as the comparable anchor — a defined compound with a reference standard that gives the same answer in any competent laboratory.

Extract Versus Satva

Worth covering here because it is a standardisation question as much as a product one.

Giloy satva is the traditional starch sediment obtained by a classical water-based process. It is starch-dominant, off-white to cream, and carries a very different marker content from a concentrated extract.

Practically:

Part: Mature Stem

The classical part is mature stem. Leaf, root and immature growth differ in composition, and giloy is harvested by cutting a climber — so mixed material is the low-effort outcome.

Require a mature-stem declaration, and limit acid-insoluble ash, which rises with woody immature material and soil carry-over.

Writing a Giloy Specification That Holds

  1. Open with species verification, not assay: "HPLC fingerprint to match authentic Tinospora cordifolia reference material; Tinospora crispa not detected. Verification reported on every batch."
  2. Name T. crispa explicitly. Excluding by naming the wanted species is weaker than excluding the unwanted one.
  3. Declare the binomial with authority — (Willd.) Hook.f. & Thomson.
  4. Specify mature stem, excluding leaf and root.
  5. State bitter principles with method, on dry basis — as a concentration measure.
  6. Require berberine by HPLC as an anchor.
  7. State extract, not satva, unambiguously.
  8. Limit acid-insoluble ash at NMT 2%.

When a Fingerprint Does Not Match

A word on how to treat a failure, because it differs from an assay deviation.

An out-of-specification assay is a conversation — natural variation, method differences, borderline results. A non-matching species fingerprint is not. There is no seasonal variation that turns T. cordifolia into something else, and no method difference that explains an absent characteristic marker profile.

Treat it as a rejection, and treat a supplier who wants to negotiate it as having answered a different question you did not ask.
