Coleus is a relief to specify after the class-assay botanicals. Forskolin is one compound, it has a reference standard, and HPLC gives a number that means the same thing in every laboratory. But that clean assay creates a false sense of simplicity, because the three things that actually go wrong with coleus are not analytical — they are the plant part, the framing of the high-assay grade, and a solvent question most buyers never look at.
What Is Coleus Forskohlii Extract?
Coleus extract is a concentrated preparation from the tuberous root of Plectranthus barbatus Andrews. The accepted botanical name changed some time ago; the trade universally retains the synonym Coleus forskohlii, and a good specification names both.
It is a member of the Lamiaceae — the mint family — native to the Indian subcontinent, where the root has long Ayurvedic use and, in some regions, is pickled and eaten. Worth noting: many ornamental garden plants are sold as "coleus" and are entirely unrelated. Name the binomial, not the common name.
The commercial identity is a single compound: forskolin (also called coleonol), a labdane diterpene.
The Good News: A Genuinely Clean Assay
After fenugreek, gymnema and grape seed, coleus is straightforward:

An independent laboratory's forskolin result should agree closely with your supplier's. If it does not, that is a genuine signal rather than a methodological artefact — which is a luxury on this ingredient list.
Trap One: The Plant Part
This is the most important line in a coleus specification.
Forskolin is confined to the tuberous root. Leaf, stem and aerial parts contain essentially none.
Now consider the harvest. Coleus is lifted whole to obtain the tuber, so at the point of collection there is a large quantity of aerial material sitting beside the root, already harvested, effectively free. Milling it in adds weight, looks entirely plausible in a brown powder, and contributes zero forskolin.
The specification line: "Part used: tuberous root only. Aerial parts (leaf, stem) absent — root-only declaration required on every batch Certificate of Analysis." This is not boilerplate on this ingredient; it is the primary raw-material control, and it is the one most likely to be quietly ignored by a supplier working to a price.
The reassuring part: HPLC catches the consequence. Aerial dilution shows up directly as a low forskolin assay, because there is nothing there to measure. So the combination of a declared root-only specification and an honest HPLC number is a strong control.
Trap Two: What a 40% Grade Actually Is
Coleus is offered at 10%, 20% and 40% forskolin. The instinct is to treat these as three strengths of one product. They are not quite.
At 40% forskolin, most of the root matrix has been removed. The material is closer to a purified compound than to a botanical extract. Three consequences:
- Regulatory classification may differ. Some markets treat highly purified plant constituents differently from traditional extracts. Confirm the position for your market and grade rather than assuming continuity from the 10%.
- Labelling should be accurate. Describing a near-purified diterpene simply as "coleus extract" is defensible but incomplete, and increasingly buyers' own customers ask.
- It handles differently. Forskolin is poorly water-soluble, so dispersibility falls as assay rises. A 40% grade disperses less readily than a 10%, despite containing four times the active. This is counter-intuitive and catches formulators.
None of this makes the 40% grade a poor product. It makes it a different product, and worth thinking about as one.
Trap Three: The Residual Solvent Line
Under-examined on coleus specifically.
Concentrating forskolin to higher assay levels commonly involves non-polar solvent steps. Hexane carries a substantially tighter acceptable residue limit than ethanol under standard solvent classifications — they are not in the same category.
A certificate reporting "residual solvents: complies" tells you nothing about which solvents were used or how much remains. Require them reported separately by name, with hexane against its own limit.
This matters more for the 20% and 40% grades than for the 10%, precisely because higher concentration means more processing.

Acid-insoluble ash deserves its limit here for a root-specific reason: a tuber grown in the ground carries soil, and inadequate washing shows up in this figure and nowhere else.
The Supply Reality
Worth setting expectations on, because coleus behaves differently from most of this category.
- It is a root crop harvested destructively. The plant is lifted to obtain the tuber — each harvest ends that plant, unlike a leaf or bark crop harvested repeatedly.
- Essentially all commercial supply originates in India. There is no meaningful origin diversification available as a risk-management tool.
- Sustained demand has pressured the cultivation base.
Practically: availability and price are more volatile than for a leaf or bark botanical, and contracting ahead of season is worth considerably more here. A lot-by-lot buyer on coleus is more exposed than on almost anything else in this range.
Category Positioning
Coleus is used predominantly in weight-management and body-composition formats — a category under sustained regulatory attention, particularly on claims and on multi-ingredient formulations.
