Coleus is, refreshingly, a botanical where a single well-chosen test closes most of the risk. Because forskolin is a defined compound assayed by HPLC, dilution of any kind shows up directly in the number — there is no cheap plant material that can quietly inflate it. That leaves exactly one sophisticated route worth defending against, and one supply-chain question the assay cannot see.
Why Coleus Is Comparatively Well Protected
The four conditions for systematic adulteration are a valuable material, a cheap substitute, an assay that cannot tell them apart, and a buyer who checks only that assay. Coleus fails the third condition, which changes everything.
As covered in our standardisation guide, forskolin is a single defined compound assayed by HPLC against a reference standard. Unlike the saponin and polyphenol class assays elsewhere in this category, there is no cheap botanical material whose constituents register as forskolin. Dilute the extract with anything and the number falls.
That is a genuinely strong position. It does not eliminate risk; it concentrates it into fewer, more specific routes.
Route 1 — Aerial Part Inclusion
The most common issue by volume, and the one driven by pure harvest economics.
Forskolin is confined to the tuberous root. Leaf, stem and aerial parts contain essentially none. And coleus is lifted whole at harvest, so aerial material is abundant, already collected and effectively free. Milled fine and blended in, it adds weight and looks entirely plausible in a brown powder.
What catches it: the HPLC assay, directly. Aerial dilution contributes no forskolin, so the number falls proportionally. There is nowhere to hide.
Supporting controls: a root-only declaration on every batch, which converts a quiet practice into a documented claim, plus TLC identity and an acid-insoluble ash limit.
Why the declaration still matters even though the assay catches it: because it changes what happens when a batch comes in low. Without a root-only clause, a supplier can present a marginal assay as natural variation. With one, they have made a specific written representation about composition — which is a different conversation, and a different liability.
Route 2 — Spent Root and Exhausted Marc
Extraction residue, dried, milled, blended back into good extract.
What catches it: the assay again. Spent root has had its forskolin removed, so the figure falls directly. On a class-assay botanical this route is usually paired with spiking to prop the number back up — but on coleus the only available spike is forskolin itself, which brings us to route 3.
Route 3 — Forskolin Isolate Spiking
The one sophisticated route, and the only one the assay alone will not expose.
A weak or heavily diluted extract is topped up with forskolin isolate from another source. The HPLC assay rises — legitimately, because there genuinely is more forskolin present. The certificate reads 10% or 20%. What is in the drum is mostly filler plus purified compound, with little actual root extract.
Note the economics: forskolin isolate is not cheap, so this route only makes sense against a substantially diluted base. It is a real practice at the margins rather than a commodity behaviour.
What catches it: the related diterpene fingerprint. Authentic coleus root extract carries a family of labdane diterpenes alongside forskolin — isoforskolin, deacetylforskolin and others — in a characteristic proportion. Added isolate brings forskolin without its companions.
The signature is unmistakable once you look for it: a dominant forskolin peak on an otherwise sparse chromatogram, with the supporting diterpenes disproportionately low or absent. It is the same logic as the flavonol ratio on ginkgo — the marker alone conceals, the profile discloses.
Route 4 — Species Confusion
More a sourcing hazard than a deliberate fraud, but worth guarding against.
Many ornamental garden plants are sold commercially as "coleus" and are botanically unrelated to Plectranthus barbatus. They contain no forskolin. A sourcing chain working from a common name rather than a binomial can genuinely go wrong.
What catches it: the assay, immediately and completely — an unrelated plant yields no forskolin at all. But prevention is better: specify the binomial, Plectranthus barbatus Andrews with the Coleus forskohlii synonym, and require it on the certificate.
Route 5 — Undeclared Carriers
Maltodextrin, starch, rice flour. Legitimate when declared.
What catches it: the assay falls, as with any dilution. Plus total and acid-insoluble ash for mineral bulking, a declared-carrier line by name and percentage, and assay on a dry, carrier-free basis.
A root-specific note on ash: acid-insoluble ash matters more here than on a leaf botanical, because a tuber grown in the ground carries soil and inadequate washing shows up in this figure and nowhere else.

Notice how much work the first row does. On coleus, an honest HPLC assay is genuinely most of the control — which is not true of most botanicals in this category.
Practical Steps for a Buyer
- Require the related diterpene profile, at least at qualification and on any source change. It is the single test the assay does not replace.
- Require the root-only declaration on every batch, in writing.
- Specify the binomial, not "coleus".
- Treat an assay discrepancy as real. Unlike class-assay botanicals, there is no method ambiguity to explain it away — this is one of the few ingredients where a divergent independent result is a genuine finding.
- Require residual solvents by name on higher-assay grades.
- Third-party verify the first batch, and put the diterpene profile in the scope alongside assay.
- Re-verify on price movements. Coleus supply is constrained and single-origin; when availability tightens, the incentive to stretch material rises with it.
