Two extracts can both be sold as “saffron extract” and be completely different products. The difference lives in which molecules are measured, how, and to what limit. This is a plain-language guide to saffron's four signature constituents and why a defensible extract is standardised to safranal and crocin by HPLC rather than colour alone.
Saffron's Four Signature Molecules
Almost everything that makes saffron valuable comes down to four related compounds concentrated in the stigma of Crocus sativus:
- Crocin — a water-soluble carotenoid glycoside. It is responsible for saffron's characteristic yellow-to-orange colour and is the marker most associated with "colour value".
- Crocetin — the carotenoid aglycone at the core of crocin; smaller and more lipophilic, and the backbone the crocin glycosides are built on.
- Picrocrocin — a bitter monoterpene glycoside responsible for saffron's taste, and the biosynthetic precursor to safranal.
- Safranal — the principal volatile, formed from picrocrocin during drying and processing. It carries saffron's distinctive aroma.
A meaningful standardisation captures at least two of these — one for colour (crocin) and one for aroma (safranal) — because they behave differently and are diluted or faked differently.
Why Colour Value Alone Is Not Enough
Traditional saffron grading (for example under ISO 3632 for the spice) relies heavily on spectrophotometric readings of colour, bitterness and aroma expressed as absorbance values. Colour is easy to measure — and, unfortunately, easy to inflate. Synthetic dyes and colour-boosting adulterants can push a colour reading upward without any real increase in genuine saffron actives.
Key point: A saffron extract quoted only on a colour figure is under-specified. Standardising to safranal and crocin by HPLC ties the number to specific molecules with specific retention times, which is far harder to fake than a single absorbance reading.
How HPLC Standardisation Works
High-performance liquid chromatography (HPLC) separates a sample into its component compounds, each appearing as a peak at a characteristic retention time. For saffron extract this gives two things at once:
- Identity — the chromatographic profile of the batch is compared against reference material. To pass, it must comply with the reference "with respect to all major peaks". A genuine saffron fingerprint is difficult to reproduce with adulterants.
- Assay — the safranal and crocin peaks are quantified against standards, giving the percentages that appear on the CoA.
SV Botanica's standard specification is Safranal not less than 0.5% w/w and Crocin not less than 2.0% w/w, both by HPLC, with identity confirmed against reference material.
Reading the Numbers on a CoA
When you see a saffron extract CoA, look for four things in the actives block:
- A stated safranal % with the method (HPLC).
- A stated crocin % with the method (HPLC).
- An identification line referencing an HPLC profile versus reference material.
- Consistency batch-to-batch — a supplier who can hold safranal and crocin within a tight band is demonstrating real process control.
Higher marker concentrations are available against buyer specification, but "higher" is only meaningful when the method is fixed. A 0.5% safranal figure by HPLC is a stronger claim than a larger number quoted with no method at all.
Why This Matters for Your Product
For cosmetic formulators, crocin drives the natural colour and antioxidant-active positioning, while safranal underpins the sensory signature. For nutraceutical formulators, the two markers are what let you make a consistent, defensible standardised-active claim.
