Almost every hibiscus specification in circulation says something like “anthocyanins 10%”. Very few say how that number was measured, what it is expressed as, or which pigments it actually sums — and on anthocyanins, unlike most botanical markers, each of those choices moves the number materially. This article unpacks the marker chemistry, the assay, the pH-dependent colour behaviour, and the specification language that separates a controlled material from a red powder.
The Marker Pair: Sambubioside Anthocyanins
Roselle's pigment fraction is dominated by two compounds. Delphinidin-3-O-sambubioside — historically called hibiscin — is the major anthocyanin, typically the large majority of the pigment load. Cyanidin-3-O-sambubioside runs second. Minor glucosides of both aglycones appear in small amounts.
What makes the pair analytically valuable is the sugar. Most commercial red botanicals — grape skin, black carrot, elderberry, purple corn — carry their anthocyanins as glucosides or acylated glucosides. Roselle attaches sambubiose, a xylosyl-glucose disaccharide, and that changes the HPLC retention behaviour enough to make genuine roselle unmistakable on a chromatogram. The sambubioside pair, in its natural proportion, is a fingerprint no cheaper pigment source reproduces.
The key insight: a total-anthocyanin figure tells you how much pigment is present; only the HPLC profile tells you whose pigment it is. On a botanical where cheaper anthocyanin sources and synthetic dyes both circulate, the profile is not a nice-to-have — it is the authenticity test. Ask for it per batch.
How Total Anthocyanins Are Actually Measured
The pH-differential method
The industry-standard assay (AOAC 2005.02 and equivalents) exploits the pigments' own pH behaviour. Anthocyanins absorb strongly around 520 nm at pH 1.0, where they sit in the coloured flavylium form, and almost not at all at pH 4.5, where they shift to a colourless hemiketal. Measuring the same sample at both pH values and taking the difference cancels out everything that absorbs at 520 nm regardless of pH — browning products, most other interferences — and isolates the genuine anthocyanin signal. It is a robust, well-validated method, and it is what stands behind our 10% specification.
The expression basis — the quiet variable
The pH-differential result must be expressed as some reference anthocyanin, because the calculation needs a molecular weight and an absorptivity coefficient. Convention is cyanidin-3-glucoside equivalents, but a result can legitimately be expressed as delphinidin-3-sambubioside instead — and because the sambubioside is a heavier molecule, the same physical sample reads higher on that basis. Two CoAs can honestly report different numbers for identical material. When comparing supplier offers, check the expression basis before comparing percentages; a specification should name it explicitly.
Where HPLC fits
HPLC quantifies each pigment individually against reference standards, giving both a total and the profile. It typically reads somewhat lower than pH differential on the same sample because it counts only the identified peaks. The practical division of labour: pH differential for the release specification, HPLC for identity and profile. A CoA carrying both is the mark of a supplier running a real laboratory.
Why the Colour Changes with pH — and Why That Is a Feature
Anthocyanins are natural pH indicators, and hibiscus formulation is mostly a story about working with that rather than against it:

Genuine roselle extract arrives with its own solution to this: the calyx's organic acids — hibiscus acid and hydroxycitric acid — pull a 1% solution to pH 2.5–3.8, squarely in the brilliant-red zone. This is why the pH line on a CoA doubles as an authenticity check, and why hibiscus performs so naturally in acidic beverages while disappointing formulators who drop it into near-neutral systems without trialling the shade first.
Stability: What Degrades the Pigments
Anthocyanins are among the less stable botanical markers, and the degradation pathways are worth knowing by name because each one maps to a formulation decision:
- Heat — thermal degradation is cumulative and accelerates sharply above ~60°C. Hot-fill processes, gummy depositing and aggressive spray drying all cost pigment.
- Light — photodegradation is real at product and warehouse scale. Amber or opaque packaging is not cosmetic for a hibiscus product.
- Ascorbic acid — counter-intuitively, vitamin C accelerates anthocyanin breakdown through a mutual-degradation mechanism. A hibiscus-plus-vitamin-C beverage needs its own shelf-life study, not an assumption.
- Metal ions — iron and aluminium complex with the pigments and shift the shade toward blue-grey; water quality and equipment contact matter.
Writing a Specification That Cannot Be Gamed
- Name the method and the basis. "Total anthocyanins NLT 10.0% w/w by UV-Vis pH differential, expressed as cyanidin-3-glucoside equivalents." Every clause closes a door.
- Require the HPLC profile. "Delphinidin- and cyanidin-3-sambubioside reported individually per batch." This is the source-authenticity control.
- Require a synthetic-colour screen. Stated as absent, with the method named. Dye spiking is the signature fraud of this material.
- Keep the pH line. 2.5–3.8 at 1% w/v. A cheap authenticity check that also predicts beverage behaviour.
