Here is the problem that defines fenugreek buying: two suppliers can both quote “50% saponins”, both be telling the truth, and be offering materially different materials. The assay is a class method, not a compound method, and it is sensitive to protocol in a way HPLC assays are not. Everything else about buying fenugreek well follows from understanding that one fact — so this guide starts there.
What Is Fenugreek Seed Extract?
Fenugreek extract is a concentrated preparation from the seed of Trigonella foenum-graecum L., a legume of the Fabaceae family grown as spice and forage for millennia. India is by a wide margin the world's largest producer. In Hindi it is methi; the Latin means roughly "Greek hay", which tells you about its forage history.
The seed carries four commercially interesting fractions, and they underpin four different grades:
- Furostanol saponins — the steroidal glycoside complex, protodioscin principal among them. The mainstream grade, typically 50%.
- 4-Hydroxyisoleucine — an unusual amino acid essentially unique to fenugreek. A specialty grade at 20%.
- Galactomannan — a viscous soluble fibre making up a large share of the seed. Sold as a fibre grade in its own right.
- Trigonelline — an alkaloid, usually a reported marker rather than a standardisation basis.
The Assay Problem — Read This Before You Compare Quotes
Furostanol saponins are not one compound. They are a family of structurally related steroidal glycosides, and the industry convention for measuring them is UV spectrophotometry after derivatisation — a method that measures the class as a whole.
That convention has three consequences a buyer must internalise:
- It is less specific than chromatography. A UV method reads a response, not an identity. Anything that produces a similar response contributes to the number.
- Protocol variation moves the result. Different laboratories using different derivatisation conditions, reference standards and wavelengths can return materially different figures on the same sample. This is not misconduct; it is the nature of the method.
- Therefore saponin percentages are not portable between suppliers unless the method matches. "50% saponins" from supplier A and "50% saponins" from supplier B are two claims measured on two rulers.
What to do about it: state the method in your specification, not just the number. Then add a chromatographic anchor: "Furostanol saponins NLT 50% w/w by UV spectrophotometry (method to be stated on CoA); protodioscin to be reported by HPLC." The HPLC line costs the supplier little and gives you a compound-specific number that is comparable across suppliers. This single addition does more for fenugreek specification quality than anything else on this page.
The Diosgenin Question
You will see fenugreek marketed on diosgenin content. Treat this carefully.
Fenugreek seed contains furostanol saponins such as protodioscin. Diosgenin is a spirostanol aglycone — it is what you get when those saponins are hydrolysed, which happens under the acid conditions used in some analytical protocols and in some processing. So a laboratory can genuinely report a diosgenin figure, but it is largely reporting the product of a conversion rather than a compound present in quantity in the native seed.
This is not a scandal; it is a well-understood analytical convention. It becomes a problem only when a diosgenin figure is presented as though it were a native content, and compared against a competitor's saponin figure as though the two were the same kind of number. If a supplier leads with diosgenin, ask two questions: is it native or hydrolysis-derived, and by what method? Note the second row. Because 4-hydroxyisoleucine is a single defined compound, it can be assayed by HPLC — which makes it a more analytically robust marker than the saponin complex. Formulators who want a chromatographically verifiable specification sometimes choose this grade for that reason alone, independent of positioning.
Two Practical Realities Buyers Underestimate
It is extremely bitter, and the aroma travels
Fenugreek's maple-bitter character comes largely from sotolone, and it survives concentration — the extract is far more intensely flavoured than the culinary seed. In a capsule this is irrelevant. In anything the consumer tastes, it dominates. Two consequences: budget for the de-bittered grade if your format is a gummy, chewable, beverage or fortified food, and bench-test sensory early, because discovering it at scale-up is expensive.
The aroma is also penetrating enough to transfer to adjacent stock in a shared warehouse. Store fenugreek away from odour-sensitive materials — this is a real and frequently learned-the-hard-way lesson.
It is a legume, and that raises an allergen question
Fenugreek belongs to the Fabaceae, and cross-reactivity with other legumes — peanut and chickpea in particular — has been reported. It is not a named allergen requiring declaration in every jurisdiction, and requirements differ by market, so whether it affects your label depends on where you sell.
The right posture is to assess it on facts rather than assume it away. Require an allergen declaration as standard documentation, and factor fenugreek into your finished-product allergen assessment — particularly if you market to consumers managing legume allergy.

The protein line is worth explaining because it is unusual. Fenugreek seed is protein-rich, and defatted seed flour — a cheap by-product — is a plausible bulking material that would raise protein while diluting saponins. A protein figure gives you a cross-check that the saponin number alone does not.
