Every botanical has adulteration risk. Fenugreek has a structural one: its headline assay measures a chemical class rather than a named compound, which means saponins from a cheaper plant can raise the number without any fenugreek being added. This is not a hypothetical vulnerability — it follows directly from the method. Here is how it works in practice, and the specification changes that close it.
The Structural Vulnerability
Adulteration appears where raw material has cost, the buyer verifies one number, and the seller's chemistry outruns the buyer's testing. Fenugreek satisfies all three, and adds a fourth condition that makes it unusually exposed.
As covered in our standardisation guide, furostanol saponins are measured by UV spectrophotometry — a method that reads a chemical response from a class of compounds rather than identifying a specific molecule. The implication is direct: anything producing a comparable response contributes to the result. Saponins are widespread in the plant kingdom, and some botanical sources are far cheaper per unit of saponin than fenugreek seed.
You do not need to posit bad actors with laboratories. You only need a method that cannot tell the difference.
Route 1 — Cross-Botanical Saponin Spiking
The defining fenugreek risk. Saponin-rich material from a cheaper source is blended into a weak fenugreek extract. The UV assay rises. The certificate reads 50%. The material contains substantially less fenugreek than declared.
What catches it: nothing in the standard assay package. This is the point. What catches it is a compound-specific HPLC anchor:
- Protodioscin by HPLC — the principal fenugreek furostanol glycoside. A spiked sample shows total saponins on specification with protodioscin disproportionately low.
- 4-Hydroxyisoleucine by HPLC — the strongest anchor available, because it is essentially unique to fenugreek. Nothing added from another botanical brings it along.
- Trigonelline by HPLC — fenugreek-characteristic and a useful secondary confirmation.
- TLC identity against reference extract, which shows the pattern rather than a single response.
The single highest-value line you can add to a fenugreek specification: "4-hydroxyisoleucine to be reported by HPLC on the Certificate of Analysis, minimum [X]% w/w." Because the compound is effectively unique to fenugreek seed, it establishes how much genuine fenugreek is in the drum in a way no saponin figure can. Route 2 — Defatted Seed Flour Sold as Extract
Fenugreek seed is protein-rich, and defatted seed flour is a cheap by-product of oil processing. Milled fine, it looks like extract, smells strongly of fenugreek — because it genuinely is fenugreek — and carries some native saponin content.
It is not extract. It is milled seed with the oil removed, at a fraction of the concentration.
What catches it: the protein figure, which is why that unusual line belongs on a fenugreek CoA. Seed flour is markedly higher in protein than a true extract, so a protein result well above the expected range for the grade is a direct indicator. Saponin assay catches gross dilution, but flour-bulked material topped up with route 1 spiking would otherwise pass.
Route 3 — Spent Seed Meal
The classic dilution: extraction residue, dried and milled, blended back. Retains the aroma, which on fenugreek is persuasive because the smell is so distinctive.
What catches it: saponin assay, since the actives are depleted — again, which is why it is normally paired with spiking rather than used alone. The pairing is the reason a single number is insufficient control on this material.
Route 4 — Undeclared Carriers and Bulking
Maltodextrin, starch and rice flour, legitimate when declared and used to make weight when not.
What catches it: total ash and acid-insoluble ash for mineral bulking; a declared-carrier line by name and percentage; and specifying the assay on a dry, carrier-free basis. On fenugreek specifically, unusually good flow and a muted aroma relative to the stated grade are informal warning signs — genuine concentrated fenugreek extract announces itself.
Route 5 — Grade Substitution Within Fenugreek
Less a fraud than a specification failure, and common. A de-bittered grade supplied against a standard saponin specification; a galactomannan fibre grade supplied where a saponin extract was meant; a 20% grade against a 50% order. These are all real fenugreek, which makes them easy to defend and easy to miss.
What catches it: naming the grade family explicitly on the purchase order and checking the assay against it on receipt. Trivial, and routinely skipped.

Practical Steps for a Buyer
- Add the 4-hydroxyisoleucine line. The highest-leverage change available on this botanical, because the compound is effectively unique to the seed.
- Never accept a saponin figure without its method. An unmethodded number cannot be compared or verified.
- Require protein. Unusual on a botanical CoA, and specifically useful here.
- Trust your nose, then test. Fenugreek's aroma is distinctive and concentration-related. Material that smells weak for its stated grade deserves a chromatogram, not a shrug.
- Third-party verify the first batch, and specify HPLC markers in the scope — an independent saponin UV result on its own reproduces the same blind spot.
- Re-qualify on any change of season or site. Fenugreek is an annual crop; supply chains shift year to year.
