Charantin sits in an awkward middle ground between the clean single-compound markers like forskolin and the wholly amorphous class assays like gymnemic acids. It is two compounds, conventionally summed, quantified against a basis that varies. Not a disaster — but enough ambiguity that a specification built on the number alone is softer than it looks.
What Charantin Actually Is
The conventional marker for bitter melon is charantin, and the name is doing more work than it appears.
Charantin is conventionally a mixture of two steroidal glucosides:
- Sitosteryl glucoside — the glucoside of β-sitosterol
- Stigmasteryl glucoside — the glucoside of stigmasterol
Two closely related sterol glucosides, historically isolated together and named as one entity.

Why the Basis Matters
Because charantin is a pair rather than a molecule, quantification requires a choice: what do we measure against, and how do we sum?
Laboratories differ on:
- Reference basis — which standard the response is quantified against.
- Whether both glucosides are resolved and summed, or one is taken as representative.
- Extraction and sample-preparation protocol, which affects recovery of sterol glucosides.
The result: the same sample can legitimately be reported at different charantin percentages by two competent laboratories.
The honest summary: a charantin figure is a reasonable relative measure within one laboratory's method and a soft absolute measure across laboratories. Not as bad as a class assay — the compounds are at least defined — but not as portable as a single-compound HPLC number. State the basis, and add an anchor.
Momordicosides: The Anchor
Momordicosides are cucurbitane-type triterpene glycosides characteristic of Momordica charantia. They are the more modern marker set and, for specification purposes, the more useful one.
The advantage is simple: individual momordicosides are defined compounds that can be quantified by HPLC against reference standards. That gives the same answer in any competent laboratory — genuinely portable in a way charantin is not.
Practical recommendation: keep charantin as the headline (the market expects it) and add a momordicoside figure as the verifiable anchor. This is the same pattern as adding 4-hydroxyisoleucine to a fenugreek saponin specification — a class or mixture number with a compound-specific number beside it.
Polypeptide-p: What the Assay Cannot See
Worth naming because it is frequently mentioned in bitter melon marketing.
Polypeptide-p is an insulin-like peptide of the fruit, of traditional and research interest. Being a peptide, it is not captured by a glucoside assay at all.
Two consequences:
- A charantin specification is silent about it entirely.
- Processing conditions that degrade peptides may reduce it while leaving charantin unchanged.
If polypeptide-p matters to your positioning, it needs its own specification and its own method — do not assume a charantin figure implies anything about it. In practice most commercial specifications sensibly do not claim it.
The Seed Question Is a Standardisation Question Too
Usually framed as a safety matter, and it is — but it has an analytical dimension.
Seed and fruit flesh have different compositions. Seed carries vicine, relevant to G6PD deficiency, and contributes different sterols and glycosides to the extract. Including seed therefore changes both the safety profile and the marker profile.
So "deseeded fruit" is not merely a safety declaration — it is part of defining what material the assay was performed on. A specification that does not state it has left the composition of the starting material open.
Bitterness as an Informal Indicator
Not a specification parameter, but genuinely useful at goods-in.
Bitter melon extract is intensely bitter, and the intensity tracks concentration. Material that tastes flat relative to its stated charantin content merits a chromatogram. It is a rougher version of the gymnema sweetness-suppression check — less definitive, but free and immediate.
Writing a Bitter Melon Specification That Holds
- Name the species by binomial — Momordica charantia L.
- Specify deseeded fruit, seed material absent, declared per batch.
- State charantin NLT X% by HPLC on dry basis, with the assay basis to be stated.
- Require a momordicoside figure as the portable anchor.
- Do not claim polypeptide-p unless you specify and test it separately.
- Set acid-insoluble ash at NMT 2%.
- Require carriers declared by name and percentage; assay on a dry, carrier-free basis.
- Screen pesticides against EU MRL — this is a vegetable crop grown to food-market practice.
When Two Laboratories Disagree
Work through in order:
- Was the assay basis the same? The most common cause on this ingredient.
- Were both glucosides resolved and summed, or one taken as representative?
- Same moisture basis?
- Do the momordicoside figures agree? If they do while charantin diverges, the difference is methodological rather than material — which is precisely why the anchor is worth having.
