The analytical problem on manjistha is unusually well defined, which is a mercy: you know exactly which species you are trying to exclude and exactly why. What makes it interesting is that the most obvious property of the material — its colour — is precisely the property that cannot help.

The second row is why this matters more than an ordinary authenticity question. Substituting one madder for another is not simply getting a cheaper plant — it is getting the plant whose preparations were withdrawn in Europe on genotoxicity grounds.

The whole genus is red. That redness is why madders were among the most economically important plants in the world for centuries.
Colour confirms the category, not the species. A rich red root powder tells you that you have a madder-type material — which is what you assumed when you placed the order. It cannot separate R. cordifolia from R. tinctorum, and buyers who assess this material by eye are performing a test that cannot fail and therefore cannot inform.
Why a General Assay Fails Too
The same structural point applies to a total-anthraquinone figure.
Both species are anthraquinone-rich red roots. A total figure is satisfied by either. This is the recurring pattern across botanical procurement: the standardisation marker and the identity question are different jobs, and a class assay answers only the first.
It is the same structure as a species-blind tannin figure on arjuna, or a bitter-principles assay on giloy. The assay is not wrong; it is simply not doing the job being asked of it.
What Works: The Fingerprint
Species verification on Rubia rests on the pattern of anthraquinones across the chromatogram rather than the amount of any one of them.
Constituents that matter in that comparison:
- Munjistin — named for manjistha itself, and a particularly useful element of the R. cordifolia profile;
- Purpurin — a principal anthraquinone of the species;
- Manjisthin — a further characteristic constituent;
- Alizarin-type constituents — shared across the genus, which is exactly why the whole pattern rather than any single peak carries the information.
Run against authentic R. cordifolia reference material, the fingerprint comparison answers a question that no single-number assay can.
Declaration Versus Result
The distinction to insist on when reading a certificate:

- Finished extract — because blending can occur after raw-material acceptance, and because the extract is what ships.
Verification at qualification only — a fingerprint run once when a supplier is approved — does not control an ongoing supply. Require it per batch.

- Aerial parts and stem milled in with root — different composition, cheaper; also caught by the part declaration and acid-insoluble ash;
- Synthetic dye added to enrich the appearance of weak material — caught by dye screening, which we regard as sensible practice on any deeply coloured botanical.
