Nearly every horny goat weed specification in circulation quotes a percentage — 10, 20, 40, 60 — and a worrying share of buyers cannot say whether that number refers to one compound or five, measured by a resolving method or a bulk one. In this botanical the difference is not academic: it is routinely worth a multiple of the price. This article unpacks the marker family, the two specification bases, and the assay language that separates a controlled material from a number on a page.
The Marker Family: Prenylated Flavonol Glycosides
Epimedium's chemistry centres on a family of flavonol glycosides carrying a prenyl group — a structural feature uncommon among dietary flavonoids and the reason the genus has its own analytical identity.
Icariin
The headline marker: a diglycoside of the prenylflavonol kaempferide skeleton, present across the commercial species and resolved as a single, well-behaved HPLC peak. When a specification says "icariin 10%", it should mean this one compound, this one peak.
Epimedins A, B and C
Close structural relatives differing in their sugar substitution. They co-occur with icariin in every genuine Epimedium extract, in ratios characteristic of the source species — epimedin C in particular runs high in some species and is a useful fingerprint peak.
Baohuoside I (icariside II)
Icariin minus one sugar. It occurs natively at modest levels, and it is also what icariin converts to under enzymatic hydrolysis — deliberately, in some processed materials, or accidentally through poor raw-material handling. An unusually high baohuoside I level in a standard extract is a processing signature worth querying.
The key insight: "total icariins" is defined arithmetic — icariin + epimedins A, B, C + baohuoside I, each resolved and summed by HPLC. It is a legitimate basis used for the 60% grade. The problem is never the basis; it is the seller who quotes the sum while letting the buyer believe they are getting the single compound. Every document should say which basis applies. On ours, it does.
Species Drives the Profile
The Chinese Pharmacopoeia admits several Epimedium species, and each carries the marker family in different proportions. E. brevicornu tends icariin-forward; E. sagittatum carries a heavier epimedin share; E. koreanum is rich in total flavonoids; E. grandiflorum — the reference species for our specification — sits in between. Two consequences follow:
- A total-icariins figure conceals species. Two 60% lots can have very different icariin/epimedin splits depending on the plant they came from.
- Profile drift usually means species drift. If your identity HPTLC or HPLC fingerprint shifts between batches while the assay holds, the most likely cause is a change — or an undeclared blend — of source species upstream.
This is why the species declaration belongs on the CoA and why the epimedin/baohuoside split should be reported per batch: together they make the profile auditable, not just the number.
How Icariin Is Actually Measured
HPLC — the method to insist on
Reversed-phase HPLC with UV detection around 270 nm resolves icariin, the epimedins and baohuoside I into individual peaks, each quantified against reference standards. Because the compounds are separated, HPLC yields both the single-marker figure and the family profile from the same run. This is the method behind every SV Botanica Epimedium CoA.
UV spectrophotometry — why it misleads here
A bulk UV method reads total absorbance and converts it to an apparent flavonoid figure. In Epimedium this is uniquely dangerous, because the epimedins absorb in the same region as icariin — so a UV "icariin" number is structurally incapable of distinguishing a single-marker extract from a total-flavonoid one. UV assay is how a 60%-total-icariins material acquires paperwork calling it high-purity icariin. If the CoA says "by UV", the number is a family total at best, whatever the label says.

Price scales with the ladder, which is precisely why the basis switch at the top matters commercially: quoted per kilogram, a mislabelled total-flavonoid lot can undercut genuine 40% icariin material while sounding superior.
Writing a Specification That Cannot Be Gamed
- Name the compound and the basis. "Icariin (single compound) NLT 10.0% w/w on dry basis" or "Total icariins (icariin + epimedins A, B, C + baohuoside I) NLT 60%". Never just "icariins".
- Name the method. "By HPLC." Without the method clause, UV is a defensible reading of your own specification — and in this botanical UV cannot answer the question.
- Require the component split. Icariin, epimedin A, B, C and baohuoside I reported individually. It is free for a supplier running a real method, and it makes both basis games and species drift visible.
- Require the species declaration. The profile only means something against a declared baseline.
- State the dry basis. Standard practice, and it closes the moisture arithmetic.
Stability and Storage
The prenylflavonoids are comparatively robust once dried — this is not a gingerol-style thermally labile system — but two practical points hold. The powder picks up moisture readily enough that an opened drum should be re-sealed with its liner intact, since moisture shifts a dry-basis assay and cakes the material. And sustained heat with residual moisture slowly hydrolyses icariin toward baohuoside I, so a warehouse holding to a single-marker specification should stay cool and dry. Store below 25°C, sealed, away from light.
