Why Grape Seed Is the Textbook Case

Grape seed extract is among the most frequently discussed adulteration targets in the botanical trade, and the reason is structural rather than moral.

The assay measures a class of compounds, not a botanical identity. Proanthocyanidins occur throughout the plant kingdom, and several cheap materials are rich in them. A class-based colourimetric assay — DMAC, Porter, vanillin — responds to proanthocyanidins from any source. It cannot tell you which plant they came from.

Combine that with grape seed extract's position as a high-volume, price-competitive commodity, and the incentive is obvious.

Route 1 — Peanut Skin

The route that matters most. Peanut skin — the thin red seed coat — is a cheap by-product of peanut processing and is genuinely rich in proanthocyanidins. Milled and blended into grape seed extract, it raises the class assay without any grape seed being added.

The consequence is not commercial. Undeclared peanut material in a finished supplement or food product means an undeclared major allergen, in a product a peanut-allergic consumer may take deliberately because it is labelled as a plant antioxidant. That is a different category of failure from a low assay.

What catches it:

If you operate a peanut-allergen programme, read this twice. Grape seed extract should be treated as a material requiring positive authenticity evidence — a fingerprint screen naming peanut markers — rather than a general supplier assurance of "no peanut in the facility". The risk here is not cross-contact in a factory. It is deliberate incorporation upstream, which no facility-level allergen control will catch.

Route 2 — Pine Bark

Pine bark extract is a legitimate commercial ingredient in its own right, proanthocyanidin-rich and available at competitive cost. Blended into grape seed extract it lifts the class assay in the same way.

No allergen consequence, but you are not receiving the specified botanical, the profile differs, and if your product literature or registration names Vitis vinifera, you have a compliance problem.

What catches it: the HPLC fingerprint again. Pine bark proanthocyanidins have a distinguishable profile — notably in taxifolin content and in the galloylation pattern, where grape seed's characteristic galloylated units are largely absent from pine bark.

Route 3 — Other Tannin Sources

Any proanthocyanidin- or tannin-rich cheap material can contribute to a class assay. Various bark, hull and pomace materials have appeared in this role.

What catches it: fingerprint comparison against authentic reference material, and the monomer profile. An extract whose class assay is on specification but whose catechin, epicatechin and gallic acid figures are disproportionate is telling you that a large share of the measured response came from somewhere other than grape seed.

Route 4 — Grape Skin Sold as Grape Seed

Within the same plant, and often not framed as adulteration at all. Grape skin extract is a genuine product — but it is anthocyanin-dominant with a very different proanthocyanidin profile, and it is not what a grape seed specification calls for.

What catches it: the profile is quite distinguishable by HPLC, and colour is a practical first-pass indicator — skin-derived material tends toward the purple-red end, seed extract toward tan and reddish-brown. Specify "seed" explicitly, not just "grape extract".

Route 5 — Total-Polyphenol Substitution

A specification failure rather than a fraud, and common. Material standardised on total polyphenols supplied against a proanthocyanidin specification. Because a Folin figure always reads higher on the same material, this looks generous and delivers less of the specified compound class.

What catches it: naming the method in your specification and requiring it on the CoA — as covered in OPC standardisation.

Note how much of this table is one instrument. On grape seed extract, HPLC fingerprinting is not a premium extra — it is the control that does almost all the work. A supplier without it cannot detect any of routes 1 through 4, whatever their certifications say.

Practical Steps for a Buyer

  1. Require an HPLC fingerprint naming peanut and pine bark markers. The single highest-value requirement on this ingredient.
  2. Require a peanut-free declaration as standard documentation. Not on request — standard, on every batch.
  3. Require the monomer profile — catechin, epicatechin, gallic acid — and look at whether the numbers are proportionate to the class assay.
  4. Name the assay method and prohibit substitution without written agreement.
  5. Specify "seed" explicitly, not "grape extract".
  6. Third-party verify the first batch with authenticity in scope. An independent OPC assay alone reproduces the blind spot exactly — the class number is not the control. Ask for a fingerprint.
  7. Re-verify periodically. Supply chains change, and this ingredient has a long history of them changing badly.
  8. Escalate internally if you run an allergen programme. This ingredient belongs on the list of materials where authenticity is an allergen control, not just a quality one.
