Almost every clove extract specification in circulation says the same thing: “eugenol NLT 60%”. Very few buyers can say what that number measures, why the method column should read GC rather than HPLC, or what a supplier can legitimately — and illegitimately — do to hit it. This article unpacks the marker chemistry, the assay method, and the specification language that separates a controlled bud extract from a number on a page.
The Marker Family: One Famous Molecule and Two Witnesses
Eugenol
Eugenol — 4-allyl-2-methoxyphenol — is a volatile phenylpropene and one of the most recognisable molecules in the botanical world: it is the smell of clove, the numbing note on the tongue, and the active that gave clove its centuries-long association with oral care. In the intact bud it constitutes the large majority of a volatile-oil fraction that can reach 15–20% of bud weight, which is why clove supports commercial extract grades of 60–80% eugenol without exotic processing.
Eugenyl acetate
The bud also carries eugenyl acetate, the acetate ester of eugenol, in meaningful proportion. This matters enormously for authenticity: clove stem and leaf oils — the cheap industrial sources of eugenol — carry almost none. An extract's eugenol-to-acetate relationship is therefore a fingerprint of what plant part it actually came from.
β-Caryophyllene
The third volatile witness is β-caryophyllene, a sesquiterpene present in characteristic proportion in genuine bud material. Together the three peaks form a profile that is easy to read on a chromatogram and hard to counterfeit convincingly.
The polyphenol fraction
Behind the volatiles sits the non-volatile fraction — gallotannins, the ellagitannin eugeniin, gallic acid and flavonoids — which is what places whole clove at the top of published ORAC rankings among common spices. The ORAC figure is a legitimate compositional descriptor of the raw material and a poor efficacy claim; regulators in the US and EU do not accept it as evidence of finished-product benefit, and serious suppliers present it accordingly.
Why the Method Column Should Say GC
GC — the method to insist on
Eugenol is volatile, thermally robust and beautifully behaved in a gas chromatograph. GC with flame-ionisation detection resolves eugenol, eugenyl acetate and β-caryophyllene into separate quantified peaks against reference standards — which is why pharmacopoeial monographs for clove materials assay by GC, and why every SV Botanica clove CoA does the same. Crucially, GC gives you the total and the profile in a single run: the assay and the authenticity check come off the same chromatogram.
UV spectrophotometry — why it invites trouble
A bulk UV method reads total absorbance and converts it to an apparent eugenol figure. It cannot distinguish bud eugenol from stem eugenol, synthetic eugenol, or any other phenolic absorbing in the same region — meaning a UV specification is satisfiable with material that contains very little clove bud at all. If a quotation is unusually cheap and the CoA says "eugenol by UV", those two facts are related.
HPLC — workable but second-best
Reversed-phase HPLC can quantify eugenol and is sometimes offered. It is not wrong, but it discards GC's advantages for a volatile analyte and often omits the acetate and caryophyllene peaks that carry the authenticity information. Accept it only with the full marker profile reported by some method.
The key insight: in most botanicals the assay tells you how much active is present and separate identity tests tell you where it came from. In clove, a properly run GC does both at once — the eugenyl acetate and caryophyllene peaks sitting next to the eugenol peak are the origin evidence. A CoA that reports only "eugenol: 65%" has thrown away the most valuable half of its own chromatogram.
Bud, Stem and Leaf: The Profile Table
The three clove-derived oils are all "genuine clove" in a loose sense, and completely different articles of commerce:

A "clove extract" standardised up with stem- or leaf-derived eugenol will show a strong eugenol peak over a hollowed-out acetate reading. It is not dangerous material — but it is not what a buyer specifying bud extract paid for, and the polyphenol fraction that motivated the purchase is diluted in proportion.
Writing a Specification That Cannot Be Gamed
A clove specification built only on a eugenol minimum leaves three doors open. Close them like this:
- Name the method. "Eugenol 60–70% w/w on dry basis, by GC." Without the method clause, UV is a defensible reading of your own specification.
- Require the marker profile. "Eugenyl acetate and β-caryophyllene reported individually per batch." This makes non-bud standardising-up visible on every CoA.
- Declare the part. "Prepared from the flower bud of Syzygium aromaticum; stem and leaf material excluded." Convert the assumption into a document.
- Set an acid-insoluble ash limit. NMT 2%. Mineral bulking in a dark brown powder shows up here and nowhere else.
Storage Is Part of the Specification
Eugenol is volatile and oxidises slowly on air exposure. An opened container left unsealed loses assay two ways at once — evaporation from the surface and oxidative darkening — and the change is one-directional. Hold stock below 25°C, sealed, out of light, and re-assay any lot that has sat open or hot before releasing it into production against a eugenol claim.
