The Acids in the Resin

The gum resin of Boswellia serrata carries a family of pentacyclic triterpene acids: α- and β-boswellic acid, their 3-O-acetyl derivatives, 11-keto-β-boswellic acid (KBA) and 3-O-acetyl-11-keto-β-boswellic acid (AKBA). AKBA is the most studied for activity and the least abundant in the native resin, typically 1% to 3% of a conventional extract.

Method 1: Total Boswellic Acids by Titration or Gravimetry

The conventional 65% grade is established by acid-base titration of the extract (an acid value converted to boswellic acid equivalents) or by gravimetric determination of the acid fraction after separation. Both count every acidic component: the six boswellic acids, other triterpene acids and any organic acid that survived processing. The result is a reliable measure of how much resin acid the extract contains, and no measure at all of which acids they are.

Method 2: Individual Boswellic Acids by HPLC

Reversed-phase HPLC with UV detection separates the six boswellic acids; the keto acids (KBA and AKBA) absorb at about 250 nm and the non-keto acids at about 210 nm, so validated methods either run two wavelengths or use a diode-array detector. A published validated method quantifies AKBA and the other five acids in Boswellia serrata extracts and commercial products against pure standards. This is the only way to state an AKBA figure, and it is also the only way to see whether a “65%” extract has the natural acid profile.

What to Specify

  1. For conventional grades: “Total boswellic acids NLT 65% by titration” plus “AKBA reported by HPLC”, so the profile is visible even when the grade is bought on the total.
  2. For enriched grades: “AKBA NLT 30% by HPLC” with the other acids reported.
  3. Residual solvent by GC where an organic solvent was used, and a solvent name on the COA.
  4. A gum-resin identity test, since other Boswellia species are traded and have different profiles.
